New Testosterone Booster Model Reveals Zinc’s Role in Prostate Cancer Risk

Scientists have developed a novel animal model demonstrating that combined treatment with zinc and testosterone significantly promotes prostate tumorigenesis in rats, offering new insights into prostate cancer development and a valuable research tool for regions with limited resources.

Prostate cancer remains a leading cause of cancer-related illness and death in men worldwide. Understanding its complex molecular and environmental causes is essential for advancing treatment. However, accessible and relevant animal models have been scarce, especially in under-resourced areas like sub-Saharan Africa, limiting research progress.

This pioneering model uses Wistar rats treated with high doses of zinc and testosterone. Alone, testosterone stimulates prostate cell proliferation without malignant changes, while high zinc levels cause epithelial dysplasia indicative of pre-cancerous lesions. Together, these agents induced aggressive tumor formation closely resembling early human prostate adenocarcinoma.

Key histopathological features observed included abnormal glandular patterns, central necrosis in tumor lesions, and stromal invasion by luminal cells. These changes mirror high-grade prostatic intraepithelial neoplasia (HGPIN) and early microinvasive adenocarcinoma, conditions recognized as precursors and early stages of prostate cancer.

Molecular analysis showed dramatic upregulation of cancer-associated genes. For instance, Tmprss2 rose nearly 790-fold, a gene frequently involved in prostate cancer progression. Akt1 increased almost 3-fold, reflecting enhanced cell survival signaling, while Pdgfrβ surged over 15-fold, indicating a pro-tumor microenvironment. Interestingly, Tp53 expression also rose, potentially indicating cellular stress responses.
