Insilico Medicine has partnered with TaiGen Biotechnology to exclusively license its AI-designed drug ISM4808 for treating chronic kidney disease (CKD)-related anaemia in Greater China. The agreement grants TaiGen and its Beijing-based subsidiary, TaiGen Biopharmaceuticals, exclusive rights to develop, commercialise, and sub-license the PHD inhibitor in the region.
The discovery and development of the ISM4808 series were recently published in the Journal of Medicinal Chemistry.
Addressing a Critical Clinical Need
CKD is linked to more than 1.5 million deaths annually. About one in seven patients develops anaemia, often due to reduced erythropoietin (EPO) levels and shorter red blood cell lifespans. Current treatments primarily relieve symptoms such as fatigue, dizziness, and shortness of breath rather than addressing the underlying cause.
ISM4808, nominated in 2022 as a potential best-in-class PHD inhibitor, targets the Nobel Prize-winning HIF‑α pathway. By stimulating EPO production and improving iron utilisation, the drug aims to enhance red blood cell formation in CKD-related anaemia. Using Insilico’s Chemistry42 engine, the molecule was designed, synthesised, and tested within 12 months prior to nomination.

Promising Preclinical Results
Preclinical studies show that ISM4808 is potent in both in vitro and in vivo assays, requiring lower effective doses in CKD rat models. The drug demonstrated oral bioavailability, favourable ADME characteristics, and promising pharmacokinetic profiles across animal models. Safety tests indicated high maximum tolerated doses and broad safety margins.
