Researchers are making significant strides toward a revolutionary oral male contraceptive pill that operates without hormones, offers full reversibility, and—most notably—has demonstrated zero side effects in its initial human trial.
The experimental drug, known as YCT-529, employs a novel contraceptive mechanism and has successfully passed a critical safety evaluation in humans. It is now poised to advance to efficacy trials, with potential availability to the public within a few years.
In the Phase Ia trial—a randomized, double-blind, placebo-controlled, single ascending dose study—16 healthy men aged 32 to 59 participated to assess the safety and tolerability of YCT-529.
After prior extensive animal testing, participants received a single oral dose ranging from 10 mg to 180 mg or a placebo under both fasted and fed conditions to evaluate absorption. They were monitored for 14 days following administration.
The findings revealed no serious side effects at any dosage level. Hormone levels remained stable, sexual desire was unaffected, and no changes occurred in inflammatory markers, mood, or key blood chemistry parameters. These results position YCT-529 as a contraceptive option free from the adverse effects often associated with female hormonal contraceptives.
Pharmacokinetic data indicated that YCT-529 has a long half-life of 51 to 76 hours, suggesting that dosing every three days could maintain effectiveness—a promising alternative to daily administration. Upcoming 28- and 90-day studies will evaluate the drug’s impact on sperm count and optimal dosing frequency.
Developed by the startup YourChoice Therapeutics, YCT-529 represents a major advancement in male contraception. Unlike hormonal male contraceptives that typically reduce testosterone and cause side effects, YCT-529 targets the retinoic acid receptor alpha (RARα), a protein essential for sperm production through the vitamin A signaling pathway.
By selectively blocking this receptor, the drug halts sperm development without disrupting sex hormones or other vitamin A functions.
Lead author Nadja Mannowetz, Chief Science Officer at YourChoice Therapeutics and a leading sperm biologist, emphasized the significance of the findings: “Our first-in-human study confirms YCT-529’s strong safety profile, with no impact on sexual desire or mood. These publications demonstrate our commitment to rigorous research, advancing the first hormone-free male contraceptive pill efficiently to meet growing demand.”
Previous animal studies in mice and primates demonstrated that YCT-529 effectively suppressed sperm production, with full fertility restoration within weeks after treatment cessation. In cynomolgus macaques, sperm counts dropped sharply within two to five weeks and normalized in about three months, again with no hormone disruption or adverse effects.
YCT-529 is currently the most promising male contraceptive candidate and, if successful in forthcoming trials, could reshape family planning dynamics by providing men with a safe, reversible, and side-effect-free contraceptive option.
