Male Birth Control Pill Clears Safety Hurdle in Health Breakthrough

by Shreeya

A major milestone in reproductive health has just been reached: the first non-hormonal male birth control pill, YCT-529, has successfully passed a Phase 1 safety trial in the United States. Developed by YourChoice Therapeutics, this oral contraceptive offers a promising new direction for men seeking more equitable options in family planning.

The drug works by blocking a vitamin A receptor in the testes, interrupting the biological process that initiates sperm production. Unlike female contraceptive pills, YCT-529 does not use hormones, a feature that significantly reduces potential side effects.

The safety trial, conducted with 16 participants, showed no significant changes in key health markers, including hormone levels, mood, sexual desire, heart rate, or inflammation. Doses of up to 180mg were well tolerated, offering early signs of promise for broader application.

Though experts like Dr. Stephanie Page, an endocrinologist at the University of Washington, advise caution in interpreting results from a small trial, she acknowledges the importance of expanding reversible male contraceptive options. “We really need more reversible contraceptive methods for men,” she said in an interview with Scientific American.

The pill requires three months of daily use to become effective and, similarly, about three months to restore fertility once discontinued—consistent with the natural sperm production cycle.

YCT-529 is one of several male contraceptives in development. Others include:

  • NES/T gel: A topical contraceptive applied to the shoulders and upper arms.
  • ADAM implant: A hydrogel-based, reversible vasectomy alternative that physically blocks sperm flow.

With nearly half of all pregnancies worldwide being unintended, the advancement of safe, effective male birth control represents a significant step forward in global reproductive health.
