Single Dose of Psilocybin Shows Long-Lasting Relief for OCD Symptoms, Study Finds

A single dose of psilocybin, the active compound in magic mushrooms, can rapidly reduce symptoms of obsessive-compulsive disorder (OCD) and provide relief that lasts for months, according to the first placebo-controlled trial examining its effects on the condition.

The study, led by Christopher Pittenger at Yale School of Medicine, involved 28 adults who had lived with OCD for an average of 20 years and had not responded to at least two conventional treatments. Participants were randomly assigned to receive either a single oral dose of psilocybin or a placebo of niacin (vitamin B3). The psilocybin dose induced a psychedelic experience, altering perception, thoughts, and emotions.

Within 48 hours, participants who received psilocybin showed an average reduction of 9.76 points in symptom severity scores, while scores for the placebo group remained largely unchanged. One week later, approximately 70% of psilocybin recipients maintained a 35% improvement, with benefits persisting at a 12-week follow-up.

“This rapid and lasting improvement is remarkable, especially in people who have not responded to traditional treatments,” said Alex Kwan of Cornell University. Experts believe psilocybin may work by increasing brain plasticity, allowing rigid, obsessive thought patterns to weaken, or by recalibrating activity between brain networks involved in self-reflection and rumination.

Despite these promising results, researchers caution that psilocybin carries potential risks. One participant in the study developed suicidal thoughts, which were managed under clinical supervision, highlighting the need for careful monitoring. Further studies are needed to determine the optimal dose, safety, and who is most likely to benefit.

David Nutt of Imperial College London noted that psychedelics like psilocybin may provide a fundamentally different approach to mental health treatment, offering hope for disorders that have been resistant to conventional therapies.

The findings underscore growing interest in psychedelics as tools for mental health, following evidence of benefits in depression, alcohol use, and other psychiatric conditions.
