Inflammation Worsens Depression Risk in Older Adults With Insomnia

Older adults who struggle with insomnia may face a significantly higher risk of depression—especially when inflammation is present, according to a new study published in JAMA Psychiatry.

Researchers at UCLA found that seniors with insomnia were three times more likely to report depressive symptoms after being exposed to an inflammation-inducing substance, compared to those without sleep problems. The study involved 160 participants aged 60 and over, with 53 identified as having insomnia.

“This study highlights a unique interaction between poor sleep and immune system responses,” said Dr. Michael Irwin, the study’s lead author and director of UCLA Health’s Cousins Center for Psychoneuroimmunology. “Insomnia not only disrupts rest but appears to leave the brain more vulnerable to inflammation-driven mood disorders.”

Participants were given an endotoxin to stimulate short-term inflammation. Among those with insomnia, feelings of depression lasted six hours or more, while healthy sleepers experienced only brief mood changes.

Researchers believe insomnia may prime brain cells to overreact to inflammatory triggers, or allow harmful biochemicals to enter the brain more easily due to sleep deprivation.

Inflammation has already been linked to chronic diseases like heart disease and cancer. As people age, inflammation tends to increase due to cellular aging, stress, illness, and other lifestyle factors—making this sleep-related vulnerability particularly concerning for seniors.

The findings underscore the potential for inflammation-targeted treatments to reduce depression in insomniacs and improve quality of life. Further research is underway to determine whether reducing inflammation could be an effective strategy to protect mental health in older adults with chronic sleep loss.
